I'd like to be able to quickly clear all flags I've set on an individual mail. I see I can right-click and under Flag, choose 'X'. But is there a way to create a one-key shortcut for that?

You coud use the predefined keyboard shortcut command + shift + L to toggle the flag on and off.

There is no way to make a one-key shortcut for anything.
You can make a custom keyboard shortcut for the Clear Flag menu item.
