I have tried all the methods suggested: Shift Safari, restarting, clearing cash and history. But I simply cannot play prime video. It started all of the sudden a couple of weeks ago.
I alway get this screen below. Is this a permanent glitch or is there a solution out there?

Since Amazon claims that they support using Safari for watching content, you will need to check with them why you are unable to use their service.

I am having the same issues on my Mac that started out of nowhere a few weeks ago. YouTube plays just fine on Safari but not Prime Video. I’ve tried the Shift - Click option, rebooting with and without power cord, rebooting router, updating video player… nothing works.
