In my Contacts, i see 3 different list. I want to have only 1 and having this to be backed up to iCloud always.

I don’t think it possible. But, I can assure you, your contacts will all be backed up and ready to go, if you ever need them.
You can contact Apple Support and ask them all the questions you want to regarding this inquiry.

A standard backup will backup all your contacts to iCloud. So you needn’t worry.

That's on setting up. I have set up but now is managing the groups. How do I merge this two groups together All Contacts (4,785) with All iCloud (4,724) so that it is backing up the 4,785 to iCloud.
Not about standard backup. But how do I merge this two groups together.
