iTunes does not back up the media (music) on an iPod.

Usually the music is already on your computer and listed in your iTunes Library, which is then synchronised with the iPod. Effectively the only method for music purchased from sources other than the iTunes Store (i.e Amazon, Bandcamp, artist websites etc.).

However, for any music purchased from the iTunes Store and downloaded directly onto the iPod, you should download the music from the store to your computer (i.e not via the iPod). I believe there is an option to automatically download those purchases to your library by going to iTunes' menu option and turning on (ticking or checking): Edit/Preferences/Downloads>Automatic Downloads>Music. Make sure to also select the Always check for available downloads as well.

I advise not relying on the iTunes Store as the backup of your music.
