After I update ipad os16.1, why I still only have mirror display when I connect my ipad to a monitor？Also I can't use Desktop‑class apps on the monitor. I have already connected mouse and keyboard. My ipad is 21pro which has M1 chip.

Apple has not, as yet, officially added external monitor support to the new iPadOS 16 Stage Manager feature.
Look for Apple to, officially, release the extended monitor support, to the iPadOS 16 Stage Manager feature, in a future iPadOS 16 update.
