Updates to the iOS should not cause any loss, but the safest option is to back up your iPod (data only) before any update.

That said, there are a couple of things to mention:

  • your music does not get backed up by an iTunes backup, mainly because it should already be on your computer, in your iTunes Library (especially since most of it is not from the iTunes Store). So if an update were to erase the music from your iPod, then a simple Sync of your iPod with its associated iTunes Library would put it back on there again
  • I'm not aware that there was ever iOS 9.0.5 for the iPod. I think it went from iOS 9.0.2, released on 30th Sept 2015, to iOS 9.1 released less than a month later, on 21st October 2015. If there is an update available for your iPod, I believe it would be straight to the last compatible version for the 5th generation model, which is iOS 9.3.5

The music in your iTunes Library (and therefore on your computer) needs to be backed up in case your computer fails. An iPod is not a suitable backup.

If your iPod is using iOS 6.1.6, that suggests that it may be a 4th generation model. However, iTunes will confirm whether or not there are any iOS updates available for that specific iPod.
