I have an external backup partition that has 300% of my system disk (750gb backup vs. 250gb disk). Time machine cannot make the backup. I read through other forums, that a backup partition with same space as the disk is enough. So, how can this be not enough?

Safe mode attempts to repair Disks & clears lots of caches & loads safe Drivers, & prevents loading of 3rd party extensions, so if Safe Mode works try again in regular boot.

finally i've tried it. for my one backup volume of 750gb this indeed worked. great thanks! But I have another volume with "only" 400gb for this 250gb disk. this didn't work even in safe boot mode. any more tipps, or do I need to reinvest to make this 400gb volume bigger?
Often a notice of Not Enough space is referring to the Internal drive where it first constructs the localsnapshot. :)

Do a Get Info on your internal drive, how much space does it have free?
Have you emptied the trash lately?
On the internal drive 64GB are available out of 250GB. Trash is empty.
