The version of the operating system has no bearing on the ports available on a Mac.
Though any iMac running Osx 10.6.8 would likely have a USB 1 or 2.0 port. You won't get USB3.2 speeds, and yes you'll likely need a USB A adapter if your flash drive is USB-C.
You can of course just look at the back of the iMac in question to see what ports it has.

Yes, an iMac from that era will have USB 2.0 support with USB-A connections, and these connections will work with a USB flash drive. It won’t get USB 3.2 speeds, however.
The first drawing below is from an iMac circa 2015, and the second drawing from circa 2009.
The iMac from 2015 has newer ports than an iMac running OS X 10.6, a version from roughly 2009.
Exactly what ports you see and what order will vary, but the shapes and the little icons shown should be similar.
The port icons shown on the first drawing are easier to see. The port icons on the second drawing are blurry in the drawing, but are shown again in blue to the left.
The only iMacs that can run Mac OS X 10.6.8 were Intel-based models released between Early 2006 and Mid 2011. They had USB-A (USB 2.0) ports.

USB-A does that.
Flip it over, try inserting it again, and when that fails flip it over again and it’ll probably insert this time. Probably. 😉
This difficulty has become somewhat of a joke, too. Newer USB (rather newer than what this iMac supports) uses a different connector (USB-C) and that newer connector usually avoids this particular flip-it-and-try-again difficulty.
Oh, and make absolutely sure it’s a USB socket you’re trying to insert into, and not some other not-USB connector.
As for the data migration, there are other ways to do backups including Time Machine, and other ways to migrate data, too.

Are you sure you are plugging it into a correct port?
The iMac should have more than one USB port, can you try the drive in a different USB port.

Don't be mad at me for saying this, but the USB-A devices can only be plugged in one way. I assume you tried flipping it over, but had to ask.
If you look inside USB-A plugs and sockets, you will see little plastic inserts. You need to plug USB-A cables in so that their plastic inserts line up with the unblocked part of USB-A sockets (and vice versa).
