What do you mean by 'downloads folder'?
Niel gave you instructions for creating a new folder, but that doesn't necessarily make it a 'downloads folder'.
That said, there are only two things 'special' about a 'Downloads folder'.
One is that the Finder has a shortcut to a folder called 'Downloads' (or whatever's appropriate in your system language of choice. As far as I'm aware this is simply a link to the folder called 'Downloads' in your home directory.
The other special thing is that web browsers such as Safari use the 'downloads folder' as the location to save files downloaded off the web. That is traditionally set to the same thing as the Finder's 'Downloads' folder, but it doesn't have to be. if you're trying to make your web browser download files to a different location, that's managed within the web browser itself.
For example, in Safari -> Settings -> General there is an option for where Safari should save files.
f you don't want the normal 'Downloads' folder, use this option to choose any other folder on disk to designate as your 'Downloads folder'.
