Are websites able to prevent us from adding screen name and password to auto-fill?
I have a website that asks me for screen name and password, but my Mac did not ask me if I want to save to auto-fill.
Then I tried to manually add, but it does not work
Any ideas?

Safe Boot, (holding Shift key down at startup), does the problem occur in Safe Mode? Could take 10 minutes or more.

Which exact OS are you running?
10.13 may need a more modern Browser than Safari on High Sierra like Firefox or Brave.
High Sierra. It has only happened twice.
