My alerts / sound effects have suddenly stopped working on my MacBook pro. It was working fine yesterday, checked system preferences, all sound settings have not been changed but no sound effects work. Any suggestions?

I'm assuming other sounds are working correctly since you specifically mention alert/sound effects.
Make sure to check the drop down list box in the Sound Effects pane to make sure the correct audio device is selected.
Try rebooting the laptop. Try a PRAM Reset (hold for at least two chimes if possible).
