Screen-Capture Not Working

Using Sierra OS, suddenly my Screen-Capture has stopped working. When using keys "Shift-Command-3 or 4", nothing happens. I've previously had no problems with this function.

Any ideas? Thank you.

iMac 27″, macOS 10.12

Thank you for your response. Since posting, I had moved on to other things, and restarted my computer. Stimulated by your answer I retried taking screenshots again, and they are now responding. However previously the screenshot was saved on the clipboard. Today it is saved on the desktop, which is OK. ?????

Is the issue isolated to certain apps? As stated in the article below, "Some apps, such as the Apple TV app, might not let you take screenshots of their windows."
