Can auto reply be turned on for messages without the driving focus?

No. It doesn’t appear to be an option.

Besides driving, in what other situations would you need auto-reply on an iPhone?
This auto-reply feature is primarily designed for when you're driving, but you can manually enable the Driving Focus mode even when you're not driving.
I got a new phone with a new number and want to set up an auto reply for incoming messages on my old phone.
