Would like to be able to control the volume and other parameters of a single layer within a patch with more then one layer, from my midi controller during a live performance.

I'm not sure what you are referring to when you say "layer". It is possible to map a physical fader to more than one MainStage fader but, were you to move a mapped fader using the MainStage interface, then the other mapped fader would also move.
Perhaps you could explain in a bit more detail your issue?
You can layer more then one instrument (sound) when creating a patch by adding instrument channel strips.
You need to go into Layout mode and put two faders on the canvas. Then assign each of them to your hardware fader. In Edit mode, you then map the faders on your strips to the faders on the canvas.
