If you are backing up to iCloud via the Backup Now, you do not lock the screen. If you want it to back up automatically while charging, the screen does need to be locked. That can be done by pressing the top button, or what I do is just let it lock itself automatically based on the time set in Settings > Display & Brightness > Auto-Lock.
