If my start up disk is almost full- how do I quickly and efficiently clean it up?

You can efficiently either delete or move to an external hard disk all the space hogging movies, videos, music, photos, etc. You need to maintain an absolute minimum of 15 GB of empty space at all times for the OS to function properly. If you allow it to go close to zero space, it will stop functioning.
