I am upgrading from iPhone 6 to the new 12 Pro. Do I need to move the SIM card or simply complete the Transfer Data steps once I power it up?

Easiest to move the SIM then go through the process for Quick Transfer.

No, if there is a new SIM in your new iPhone from your carrier you can activate it.
